Organelle Movement in Actinophrys sol and Its Inhibition by Cytochalasin B

چکیده
Movement of extrusomes in the heliozoon Actinophrys sol was characterized at surfaces of the cell body and giant food vacuoles where microtubules are absent. Extrusomes moved in a saltatory manner at an average velocity of 0.5 μms-1. The highest velocity observed was 2.1 μms-1. Cytochalasin B (50 μg/ml) strongly inhibited extrusome movement at the surfaces of newly-formed food vacuoles, suggesting that the actomyosin system is involved in the organelle transport in Actinophrys.

Organelle movements in the wild type and wall-less fz;sg;os-1 mutants of Neurospora crassa are mediated by cytoplasmic microtubules.
The cellular basis of organelle transport in filamentous fungi is still unresolved. Here we have studied the intracellular movement of mitochondria and other organelles in the fungus Neurospora crassa. Four different model systems were employed: hyphae, protoplasts, a cell wallless mutant, and experimentally generated small, flattened cell fragments of the mutant cells. Organelle movements were...

The apical organelles in apicomplexan parasites are characteristic secretory vesicles containing complex mixtures of molecules. While apical organelle discharge has been demonstrated to be involved in the cellular invasion of some apicomplexan parasites, including Toxoplasma gondii and Plasmodium spp., the mechanisms of apical organelle discharge by Cryptosporidium parvum sporozoites and its ro...

Exocytosis of extrusomes, secretory granules found in protozoa, is involved in prey capture by the heliozoon Actinophrys sol. Here we show that extracellular Ca 2+ is necessary for exocytosis and prey capture in A. sol. We found that A. sol could not capture prey cells in a Ca 2+-free solution. L-type Ca 2+ channel blockers and a calmodulin antagonist also inhibited the capture of prey. These r...
متن کاملInfluence of colchicine and cytochalasin B on pinocytosis, phagocytosis, and antibody-dependent cell-mediated cytotoxicity.
The cytoskeleton is an organelle of eukaryotic cells engaged in organization and external and internal cell movements, e.g., endocytosis. Our previous experiments proved that colchicine (an antitubular drug) and cytochalasin B (cyt B, an antimicrofilament agent) have significant influence on the development of the process [8, 9]. Reports on the inhibiting effect of the development of antibody-d...
متن کاملP-75: The Effect of Hydrostatic Pressure in The Presence of Different Concentrations of Extracellular Calcium, Ethanol and Cytochalasin B on Parthenogenetic Activation of Mouse oocytes
Background: The parthenogenetic of oocytes is important in cloning research, as artificial activation of oocytes is an essential component of nucleus transfer protocols. Hydrostatic pressure can act as a mechanical stimulator that rearranges egg contents. Ethanol promotes a single intracellular Ca2+ increase of greater and longer amplitude than the initial increase observed at fertilization. In...
